Essay

The thesis is devoted to the study of viral diseases mushrooms, improvement of methods of identification dsRNA from fruiting bodies of fungi mushrooms and the development of molecular-biological test systems mycoviruses in greenhouses. We performed primary screening of mushroom samples grown in a tunnel to the virus infection using the methods of visual diagnostics of fruiting bodies and mycelia and electron microscopy. Studied the prevalence of viral diseases of mushroom in the Kiev region. Adapted and improved methods of isolation, purification and identification of viral double-stranded RNA (dsRNA) in mushrooms. Optimized method for the extraction and purification of dsRNA from the MVX (Mushroom Virus X) mushroom crops. A design of primers and optimized PCR conditions for the diagnosis and identification of MVX. Developed a multiplex PCR diagnostic test system for detection of pathogenic mycovirus MVX (. Optimized composition of substrates and culture media for the cultivation of mushroom in a tunnel. Improved production of biotechnology in industrial production of mycelium mushroom based on without virus. As a result of research is recommended mandatory control planting material fungi (mycelium) in the presence of viral pathogens. It is recommended for asymptomatic mushrooms, which in turn may be exposed to a virus in a latent (asymptomatic) form. The latter may be a hidden reason for the emergence of mixed infection, when infected with a virus, mushroom crops are susceptible to diseases caused by bacteria and other pathogens, resulting in loss of productivity and product quality. Tested system of diagnosis and identification dsRNA fragments allows for rapid laboratory diagnosis of virus MVX mushrooms.
