Essay

The object of the research was the biological action of aqueous-saline extracts of cryopreserved fragments of pigs' heart and piglets' heart. The research aim was to determine the effect of cryopreservation conditions of pigs' and piglets' heart fragments onto the composition derived from them the aqueous-saline extracts and the effect of these extracts on normal rats and rats with myocardial ischemia. In the work the following methods were used: spectrophotometry, spectrofluorimetry, gel-penetrating chromatography, matrix-assisted laser desorption/ionization, histological, immunohistochemical, light microscopy, electrocardiography and methods of statistical processing of the measurement results. Devices which were used in the work: ultraviolet detector to chromatography LKB-2238 Uvicord S11 (LKB-Produkter AB, Sweden), time-of-flight mass-spectrometer MALDI MS Autoflex II (Bruker Daltoniks GmbH, Germany), spectrofluorimeter Cary Eclipse (Varian, Australia), hardware-software system for registration of electrocardiograms "Poly-Spectrum" (Neurosoft, Russia). The results. It was found that yielding of the substances of peptidic nature into solution is higher from the fragments of heart cryopreserved under protection of polyethylene oxide with m. w. 1,500. It was established that components of the studied extracts bind to albumin of blood serum, and this binding can be considered as one of the specific mechanisms of transport of biologically active substances to appropriate organs and tissues. Obtained data testify that the condition and ratio of regulatory systems of the heart does not change when injected to animals the extract of the cryopreserved fragments of pigs' heart or piglets' heart. It was established that the injections of the extracts from cryopreserved fragments of piglets' heart have positive effect on the condition of heart muscle of the rats with myocardial ischemia, manifesting the normalisation of electrophysiological indices of heart functions. Novelty. For the first time defined the composition of the extracts of cryopreserved fragments of pigs' heart and extracts of cryopreserved fragments of piglets' heart in dependence upon the type of cryoprotectant and its concentration. It was shown that the injections of the extracts of cryopreserved fragments of piglets' heart during 2 months to animals with myocardial ischemia normalize the blood supply of heart muscle. It was established that the introduction of extracts of cryopreserved fragments of heart causes to increasing the proliferative activity of heart cells.
