Essay

The object of the study - the influence of culture, hypothermic storage in mannitol containing media and cryopreservation on the structural and functional safety of pancreatic microfragments and islet of neonatal piglets under transplantation to experimental animals. The aim of the work - to study the influence of culture, hypothermic storage and cryopreservation of pancreatic tissue of neonatal piglets on its structural and functional properties as well as the study of the level of glycemia and level of reparative processes in the animals with experimental type 1 diabetes after heterotopic transplantation of pancreatic tissue or islets. Methods: culturing, cryopreservation, transplantation, enzyme immunoassay, immunohistochemistry and morphometric analysis, light and fluorescence microscopy. It is found that "HTK" solution can be used to store the tissue prior to transplantation. Cryopreservation with 10% DMSO and cooling rate of 1°C/min allows to achieve high structural integrity of pancreatic microfragments. Subcutaneous transplantation of pancreatic tissue to the animals with type 1 diabetes leads to the activation of reparative processes in the recipient's.
