Essay

The thesis is devoted to investigation of the role of nitric oxide and its derivate - peroxynitrite - in hypoxia-induced damage of mice dorsal root ganglion neurons. The data obtaned indicate that low-voltage activated (LVA) and high-voltage activated (HVA) calcium currents (ICa) are resistant to hypoxia in 80% of neurons. Hypoxia reduced both ICa LVA and HVA in 20% neurons only. At the same time, rotenon caused irreversible decrease of ICa in all kinds of investigated neurons. Authentic NO reduced both resistant and sensіtive to hypoxia LVA and HVA calciun currents. This inhibitory action was shown to be mediated substantially by an increase in cGMP level. Peroxynitrite irreversibly blocked both types of calcium currents. Effects of direct NO-donors - sodium nitroprusside and SIN-1 - on calcium currents was shown to be different from effect of authentic NO.
