Research object - the impact of cryopreservation and induction of donor-specific tolerance on structural and functional preservation of endocrine glands tissue fragments (thyroid, testes, adrenal, ovary) after transplantation to experimental animals, as well as the expression of immune proteasomes and content of regulatory T cells CD4+CD25+Foxp3+ in recipients. Aim - to examine the effect of cryopreservation and induction of donor-specific tolerance on structural and functional preservation of endocrine glands tissue grafts, and to identify biological markers of transplant tolerance in recipients. Research methods - cryopreservation, cultivation, transplantation, radioimmunoassay, flow cytometry, spectroscopy, light and fluorescence microscopy, histological analysis, morphometric analysis, immunohistochemical analysis, Western blotting, statistical analysis of results. For the first time the efficiency of cryopreservation and induction of donor-specific tolerance to achieve a high structural and functional preservation of endocrine tissues after transplantation was established as well as the expression of imunoproteasome subunits LMP2/LMP7 was proposed as a biological marker of transplantational tolerance.
