Essay

In the dissertation, an important scientific and technical task is theoretically grounded and practically solved this consisting in the development of the scientific foundations of biotechnological approaches to preservation of the activity of starter microbiota for dairy products of a traditional assortment range and functional action. A comprehensive approach to control the growth of inhibitors of fermented microbiota has been developed, including an analysis of the content of technologically harmful microflora, chemical inhibitors, and bacteriophages. From the standpoint of industrial biotechnology, an “Instruction for the Organization of Industrial Microbiological Control at Dairy Enterprises” has been developed, which defines the rules, indicators for controlling the technological process of dairy production, offers appropriate methods and analysis tools taking into account the requirements of the regulatory framework and the frequency of control, both fermented and fermented extraneous microflora, inhibitory substances and bacteriophages. For the first time, critical control point schemes were developed with enhanced control according to the technological stages of dairy production. Technology has been developed for the production of test cultures for the selective monitoring of inhibitors of fermented microbiota: inhibitory substances in milk raw materials and bacteriophages of lactic acid bacteria. Selection methods and the specificity of the use of test cultures for the detection of inhibitors of fermented microbiota have been established. The conditions of modern methods for detecting phages of lactic acid bacteria have been substantiated and established: using transmission electron microscopy and PCR, multiplex-PCR. The structure of the population of bacteriophages circulating in milk processing plants of Ukraine was investigated. A systematic collection of phages of lactic acid bacteria of various taxonomic groups has been created: L. lactis ssp. lactis, L. lactis ssp. cremoris, L. lactis ssp. lactis biovar diacetylactis, En. faecium, Lb. acidophilus та S. thermophilus, isolated at the factories of Ukraine, which formed the basis for the development of modern biotechnological approaches for the identification, inactivation of phages and their use for the selection of resistant cultures.
