Essay

Erythrocytes of the human donor's blood are the object of the study. The aim of the investigation is to study the effect of different physical and chemical factors on the structural and functional state of erythrocyte cytoplasm and their membrane permeability for cryoprotectants. The main investigation methods and apparatuses: the EPR spectrometers ("Bruker ER-100D" (Germany)), the NMR "Tesla BS 567A" with the 100 MHz wor-king frequency (Czechoslovakia), the "SRECORD PAY UNICAM SP-8000" spectrophotometer (Great Britain) or "SRECORD UV VIS" (Germany), the FK-56M colorimeter (USSR), the UOP-8 programmable freeze, the OMKA 1Z osmometer. Scientific novelty. Systematic investigations of erythrocyte membrane permeability for different cryoprotectants has been conducted for the first time, the effect of an intracellular protein concentration on the redistribution of different substances between erythrocytes and their environment and the rate of glycerol permeability has been studied. Main results: It has been established, that cryoprotectants by the permeability coefficient size form a row, where the cryoprotectant position is determed by the complex of all its physical and chemical properties and the concentration of an intracellular protein affects redistribution of different substances between erythrocytes and their environment and the rate of glycerol penetrating. A theoretical approach for the creation of economic and less labor-consuming cryopreservatives for the cellular suspension cryopreservation, has been elaborated. Practical significance: economic, effective labor-consuming methods of human erythrocyte cryopreservation at -196 and -85... -90 оC has been created. A simple method of the initiation of water crystallization in the container for the prevention of a negative cooling effect has been suggested. The phenomenon of the increase of the instinction of methemoglobin of chromophoric group at the presence of erythrocytes was used to study the effect of different substances on the erythrocyte membrane
