Essay

The dissertation presents the results of pathological-anatomical, histological, histochemical, toxicological, and chromatographic studies of the organs and tissues from 19 dogs of different breeds and ages that died due to acute spontaneous isoniazid poisoning. The following research was conducted and the results analyzed: pathological-anatomical changes (macroscopic changes detection), histological changes (evaluating microstructural changes using hematoxylin and eosin staining of histological preparations of the stomach, liver, pancreas, kidneys, heart, lungs, brain, spleen, and lymph nodes), histochemical studies (detection of ribonucleic acid by Brachet method; glycoproteins – MacManus’ PAS reaction; hemosiderin - Perl's method; chromaffin substance in neurons – Nissl staining), chromogenic reactions (extraction studies from the liver and kidneys), and liquid chromatography (identification and quantitative determination of isoniazid content in the stomach) using a chromatograph with a dual mass spectrometric detector Waters XEVO TQ-S Micra in the chemical-toxicological department of the State Research Institute for Laboratory Diagnostics, Veterinary and Sanitary Examination (Kyiv). These methods allowed to establish the features of pathological and anatomical changes depending on the duration of clinical symptoms in dogs and thus diagnose isoniazid poisoning.
