Formalin-water extracted antigens, whole cell sonicated extracts were used as leptospiral antigens for ELISA.The reactivity spectrum of the antigenic preparations was determined by detecting antibodies in field and immune sera. The antigen adsorption onto the polystirol surface of the plates was carried out at room temperature till full evaporation of the liquid, by overnight incubation of the sensitised plates at 40C, or by a two-hour incubation at 370C. For antibody detection in bovine and swine sera immunoperoxidase conjugates were used, whereas for canine and rabbit sera ProteinA-peroxidase conjugates were used. OPD was used as a peroxidase substrate for automated reading and 5-amino-2-salicylic acid for eye reading of the results.
