Through the restriction analysis, dot, blot-hybridization the genome rearrangements at DNA sequence level in cultured in vitro R.serpentina cells have been demonstrated. Correla-tion of the changes' magnitude with the duration of the plant cells culturing in vitro was estab-lished. Occasionally the genome changes to occur in plant cells during culturing in vitro by their range may exceed the interspecies variation to arise during evolution upon origin of the individual species. It was established that liable to changes were the specific fractions of the repeated sequences such as the moderate repeats, inverted reiterations (immediately reassoci-ating), DNAse-resistant fraction, individual fragments and certain genes. Plant genome in con-ditions in vitro was found to exhibit various DNA rearrangement patterns, namely, changes in the copy number of some fractions and fragments, mutations and micromutations that result in RFLP both in the individual fragments of the genome DNA and in some structural genes . There also may occur changes in the methylation pattern for the cytosine residues within CG duplexes of some genes. It was revealed that the genome changes display non-random pattern. Parallel-ism between the genome changes in culture in vitro and interspecies variation among intact plants, members of Rauwolfia genus, was brought out. Speculation that some genome changes in cultured cells may occur according to the law of the homology raws for hereditary variability by M.I.Vavilov.
