Essay

The object - rabies virus, enzyme-linked immunoassay. The aim - to work out an rabies specific antibodies test-system based upon enzyme-linked immunoassay. The methods - virological, immunological, hematological, biochemical and statistical. The novelty - for the first time diagnostic highly specific hyperimmune rabies antisera was obtained by means of novel immunization schedule, which based upon fivefold injection of concentrated cultural rabies vaccine - CoCAV to rabbits on the 1st, 3rd, 7th, 14th and 30th days with addition of adjuvant ISA 25 on the 1st and 14th days of immunization course. The method of rabies virus concentration with polyethyleneimine was offered. Diagnostic enzyme-linked immunoassay was elaborated on the base of indirect ELIZA with involving of recombinant staphylococcal A-protein conjugate. The test-system allows to detect rabies specific antibodies in sera of different mammalian species, and put the results into International Units (IU). The novelty is confirmed by DeclarativePatent of Ukraine № 64544 A 7 G01/N 33/00.Field - veterinary medicine.
