Essay

In dissertation the results of research of karyotype abnormalities, fusion genes, clonal rearrangements of Ig genes, disturbances in microsatellite DNA are presented comparing with the indexes of acute leukemia flowing in children. It was demonstrated that the presence of cells with relative clones is predictive unfavorable sign of acute leukemia flowing, and PCR-positive patients according to the genes AML1/ETO and TEL/AML1 have the best therapy response. It was revealed that clonal rearrangements of Ig genes and disturbances in microsatellite loci D9S171 and D12S98 can be used as molecular marker of the tumor clone in children with acute lymphoblastic leukemia. It was found that the estimation of minimal residual disease, based on the investigation of fusion transcripts and clonal rearrangements of Ig genes, allows to select patients with the higher risk of development of acute leukemia early relapse. On the basis of these data the spectrum of genetic criteria of disease flowing prediction was extended. The optimum chart of genetic abnormalities researches in children with acute leukemia was worked out.
