Essay

Object: complexes of gramicidin S with platelets and erythrocyte membranes. Purpose: elucidation of regularities of platelets aggregation and erythrocyte hemolysis under the influence of different factors that change chemical composition and structural condition of membranes. Methods: optical method of registration of platelets aggregation and erythrocyte hemolysis, spectrophotometry, photometry, fluorescent microscopy, chromatography, determination of POL products, statistical analysis methods. Results, novelty: it was at first shown that interaction of gramicidin S with platelets leads to the shape change which analogous to that takes place under the influence of known aggregation inductors; gramicidin S destructs platelets aggregates forming under the influence of aggregation inductors not destructing cell membrane. Decrease of lipids ranging in platelets and erythrocyte membrane under their ?-irradiation or lipid peroxidation results in the facilitation of gramicidin S integration into membrane and simultaneously leads to the decrease of binding strength of antibiotic with it. Degree and velocity of platelets disaggregation by gramicidin S are the most in the region of membranes structural phase conversions. It was shown at first that gramicidin S biological activity was dependent on cholesterol content and cholesterol/phospholipids ratio in membranes. Field of application: human physiology.
