Essay

The subject of study is DNA of calli tissues and intact plants of different maize lines. The aim is to reveal the peculiarities of genome variability into maize tissue culture depending on the genotype and culturing duration. The methods of study are plant tissue culture, DNA isolation, RAPD- and SSR-PCR, DNA electrophoresis into agarose and acrylamide gels, Southern hybridization, statistical methods. Every line involved was recorded to show little number of the genome rearrangements in conditions in vitro. A bulk of rearrangements was observed at the early steps of culturing (up to two months). Considerable proportion of rearrangements to be registered in two months old callus tissues was found to be of unstable nature being not observable following four months long cultivation. At least subset of the changes revealed was shown to exhibit not qualitative (amplicon presence/absence within the fragments spectrum) but quantitative character (copy number variation). Field of use is plant breeding.
