Essay

Dissertation devoted to the study of cultural-biochemical, microbiological, serological and molecular - biological investigations. It was chosen and characterized strain Erysipelothrix rhusiopathiae М-2 VК for producing of the inactivated vaccine against swine erysipelas, optimized nutrient media and terms of cultivation for obtaining maximal bacterial biomass. It was offered the method and the order of the inactivation of erysipelas bacteria and experimentally substantiated using of adjuvant "Montanide ISA 25". The immunity of the Erysipelothrix rhusiopathiae М-2 VК strain, which was characterized as mutant strain with insertion in the main protective gene spa A, was confirmed in the experiments carried out on laboratory mouse and swine in pig farm condition. It was investigated what strain Erysipelothrix rhusiopathiae М-2 VК had insertion in spaA gene with size near 400 nucleotide pair. Erysipelothrix rhusiopathiae strains ВР-2 var. ІVМ, 149, Ш and М-2 VК. Experimental emulsion-vaccinein 14 days after single shot injection (dose per mouse - 2 billion microbial cells) provided 92 % protection of vaccinated mouse and 75 % pigs in six months after single shot vaccination with vaccine, contained 8 billion inactivated microbial cells.
