Bublyk O. Peculiarities of somaclonal variability of Ungernіa vіctorіs Vved. ex Artjushenko

The subject is genome variability in plant tissue and cell culture. The aim is to study genome variability of U. vіctorіs tissue culture under various conditions and duration of cultivation, to compare the level and peculiarities of U. vіctorіs genome variation in nature and in tissue culture. The methods are plant tissue culture, RAPD-analysis, Southern blot hybridization, cytogenetical analysis. Long-term maintenance of U. vіctorіs tissue culture was found to be accompanied with the genome changes at chromosome and DNA levels. Molecular genetical changes pattern in most cases reproduced differences between plants from natural population, while being by the level almost twice orders of magnitude lower. The influence of nutrient medium composition, conditions of cultivation, type of callus, culture period duration on the level and peculiarities of somaclonal variation were found.
