The thesis is devoted to the improvement of porcine enzootic pneumonia (PEP) diagnostic tools by the introduction of molecular genetic methods based on PCR and using recombinant antigens for serology tests. The Mhp366 and Mhp377 membrane lipoproteins of M. hyopneumoniae, which have immunological importance, were characterized. Their synthetic copies in composition with GST fusion proteins were obtained using recombinant DNA technology. It was demonstrated the high species-specificity of obtained fusion proteins and a practicability for their using as antigen for the head screening concerning mycoplasma infection. The construction method of the PEP molecular diagnostics based on the PCR was created. The diagnostic kit "SuiDNAtest M. hyo" for the M. hyopneumoniae DNA detection in clinical material was designed. The combination with the commercial ELISA and viruses detection tools allows conducting monitoring and determining the mycoplasma infection in head. For the first time molecular-phylogenetic studies of Ukrainian isolates of M. hyopneumoniae were carried out.
