Essay

Theoretical study of the structure of the glycoprotein B precursor has been carried out, its variability, secondary structure have been determined, and the theoretical spatial model has been obtained. There has been developed the method of indicating DNA viruses of the subgenus alfa Herpesvirinae genus Mardivirus and differentiation of Marek's disease agent Gallid herpesvirus 2 using multiplexed PCR, which showed intraspecific specificity. There has been constructed plasmid pUC_gB, able for the expression of glycoprotein B in eukaryotic cells, its authenticity was proved by PCR, RFLP and electrophoretic analysis. There has been determined that developed on the base of plasmid pUC_gB liposomal form of DNA vaccine has distinct protective properties, it is nontoxic and harmless for one day old chickens. As a result of sequencing plasmid pUC_gB there has been determined the structure of gene gB of the strain JM-UA Gallid herpesvirus 2, and its phylogenetic study has been carried out. Based on experimental studies on the construction of vector molecules, development of medical form, study of non-toxicity, harmlessness and protective properties there have been laid down general regulations for the quality control of DNA vaccines.
