Essay

The dissertation is devoted to the experimental substantiation of appropriateness of usage thiol-containing drug acetylcysteine with the medical purpose on the model of closed traumatic brain injury. In a series of pharmacometric studies it was proved that acetylcysteine shows cerebroprotective effect in a dose 137,1 mg/kg as 2% aqueous solution in 30 min after causing of mechanical brain injury and in dose 123,4 mg/kg in 6 h 27 min afterwards. It has been proven that pharmacotherapeutic efficacy of acetylcysteine is realized by preventing of free-radical reactions branching and consequently, oxidative stress, which develops on background of traumatic brain damage. Administration of acetylcysteine to injured animals normalizes the main indexes of energy and carbohydrate metabolism, increases physical performance and dynamic endurance and also stabilizes parameters of acid-base balance and oxygen transporting system of blood of rats with closed craniocerebral injury. Pharmacotherapeutic use of acetylcysteine helps to accelerate the detoxification from endotoxins, circulating in the blood of rats with closed cerebral injury.
