Essay

The dissertation thesis includes the theoretical and experimental substantiation of the improving of the laboratory diagnostics of Fusobacteriosis. Indication, identification and selection have been made of the PRODUCTIVE STRAINS OF Fusobacterium necrophorum, has been STUDIED VIRULENCE AND MOLECULAR GENETIC PROPERTIES of the CIRCULATING EPIZOOTIC STRAINS IN UKRAINE BY CHOOSING THE OPTIMAL CULTURE MEDIA FOR THEIR CULTIVATION. THE RESULTS OF THE DEVELOPMENT OF THE TEST-SYSTEM "ELISA-Fusobacteriosis" for the INVESTIGATION OF THE SPECIFIC ANTIBODIES IN LABORATORY ANIMALS AND CATTLE AFTER THE IMMUNIZATION BY VACCINE against Fusobacteriosis have been PRESENTED AND its EFFECTIVENESS has been PROVED IN INTER-LABORATORY TRIALS. For the first time it has been DEVELOPED the PROCEDURES OF RECEIVING SPECIFIC INACTIVATED ANTIGENS FROM SURFACE-ACTIVE peptides of the CELL WALL OF HIGHLY PATHOGENIC STRAINS F. necrophorum and it has been studied their immunogenic and antigenic activity IN RABBITS AND WHITE INBRED MICE. It has been developed the new procedure of immunization of donor animals and positive specific sera against Fusobacteriosis with the titers of antibodies 1:5120 - 1:10240 have been received. As a solid carrier in the ELISA polystyrene micropanels of the company "Sarstedt" (Germany), which were used adsorbed by the purified and concentrated antigen from the cell wall polypeptides of F. necrophorum with the concentration of 10 ?g/cm3. ADSORPTION has been CARRIED OUT AT THE TEMPERATURE OF 4 °C FOR 20 HOURS. DURATION OF INCUBATION WITH SERA ADSORBED with ANTIGEN has been AT THE TEMPERATURE OF 37 °C FOR 60 MINUTES. THE WORKING CONJUGATE'S DILUTION FROM PROTEIN A of STAPHYLOCCOCUS AUREUS LABELED by HORSERADISH PEROXIDASE was 1:4 000. The results of the researches received in the ELISA, have been COMPARED WITH THE RESULTS OBTAINED IN THE AGGLUTINATION TEST AND HEMAGGLUTINATION INHIBITION TEST. THE TECHNOLOGY OF PRODUCTION AND CONTROL of the TEST-SYSTEM "ELISA-Fusobacteriosis" has been developed.
