Essay

The object is a detection of latent virus infection in healed potato lines through EIA and RT-PCR methods; variability of healed potato lines in the application of thermo and chemotherapy; causal relationships of variability and feasibility of PCR analysis application in the production of quality seed potatoes. The aim is to assess and allocate productive meristem lines to reproduce pre-basic seed potatoes by using molecular and genetic analysis methods. The methods are molecular and genetic, laboratory, field, visual, measurement and weighting, statistical, settlement and comparative ones. For the first time in Ukraine the test system based on RT-PCR for the diagnostics of potato spindle tuber viroid and X-, Y-, S-, L-, M- viruses; molecular and genetic methods (RAPD- and ISSR-PCR) of identification of potato lines' variability following the healing of the varieties with biotechnological methods have been developed for seed potato system. Using modern molecular and genetic methods in biotechnology makes it possible to shorten for a year the test term of the lines under the complex of economically valuable performance in the field. The main research results were introduced in 2010 and 2013 at the Institute for Potato Research, Institute of Agricultural Microbiology and Agricultural Production of NAAS and the farm enterprise "Adelaida" (Kherson). Field of application is agricultural production.
