Dissertation focuses on the physiological mechanisms of impact of pyrrolidine-2-carboxylic acid derivatives in gastric mucosa under the experimental ulcers. The experiments revealed that the test compound sodium 2- (2-hydroxyphenoxy) acetyl) -L-prolinate had pronounced antiulcer effect in models of experimental ulcer caused by stress, ethanol and nonsteroidal anti-inflammatory drugs (indomethacin and aspirin). Analysis of the mechanisms of antiulcer action of sodium 2- (2-hydroxyphenoxy) acetyl) -L-prolinate showed that such properties of compound is associated with a decrease in the secretion of hydrochloric acid in the stomach by partial inhibition of H+/K+-ATPase activity, reduction of basal and stimulated secretion of pepsin, acceleration of mucous epithelial barrier recovery, that was evident by lowering of oxyproline, fucose and hexuronic acids content in gastric mucosa, increased secretion of prostaglandin E2. The level of proinflammatory cytokines (interleukin-1b, 12B p40, tumor necrosis factor-a, interferon-g) in serum of rats was decreased under the conditions of therapeutic administration of test compound after ulcer modeling, indicating a reduction of the inflammation intensity in conditions of the gastric ulceration.
