Essay

The dissertation is devoted to the scientific and experimental substantiation of the manufacture of an inactivated vaccine against infectious rhinotracheitis. The research questions of the cultural properties of the infectious rhinotracheitis virus strain «BM» have been highlighted and the optimization of its accumulation. The technology of infectious rhinotracheitis virus has been developed accumulation in the roller culture of MDBK transfected cells with high biological activity (8.63±0.12 lg TCD5o/cm³). The method for the hyperimmunization of rabbits has been developed to obtain a specific high-level hyperimmune serum (10.04 log₂). Ethyleneimine has been used for the inactivation at concentration of 0.01 %, which reduces the inactivation process to 24 the conducted comparative study hours. The conducted comparative study of a number of substances as adjuvants showed that the most active immune response to the administration of the inactivated vaccine against infectious bovine rhinotracheitis was observed in animals using a mixture of highly purified mineral oil and aerosol as an adjuvant. It has been established that the developed vaccine was manufactured in accordance with the requirements for inactivated vaccines for veterinary medicine. The study of immunogenicity has shown the effectiveness of the developed inactivated vaccine against infectious bovine rhinotracheitis. It allows you to create livestock immunity for 28 days after vaccination at the level 9–11 log₂. It has been shown that the application of the developed vaccine preparation in 150 days resulted in a level of specific antibodies in the range of 10.80–10.88 log₂. The duration of this specific immunity in animals is at least 9 months (8.0–8.2 log₂). The effectiveness of the vaccination of single cows has been shown. Because in the future when the colostrum was given to newborn calves, they formed colostral immunity to IBR, which predetermines the protection of animals before the first vaccination.
