The thesis is devoted to the obtaining biotechnological plants of bread wheat by Agrobacterium-mediated transformation with the gene of ornithine-δ-aminotransferase Medicago truncatula and the study of their morphological, physiological, molecular-genetic and cytological characteristics. During the research, an in vitro biotechnological system for Agrobacterium-mediated transformation of bread wheat was developed and the process of obtaining plants in the culture of shoots apical meristems was improved. In order to find the most suitable genotype for the Agrobacterium-mediated transformation, the processes of morphogenesis in the culture of three different types of explants (the shoots apical meristems of of 3-day seedlings, immature and mature embryos) of 15 modern high-yielding wheat varieties were analyzed. The stimulating effect of the antibiotic cefotaxime on the regenerative capacity of wheat callus cultures, obtained from explants of immature embryos was shown. Conditions for conducting Agrobacterium-mediated transformation of morphogenic calluses of wheat varieties have been optimized. It was performed Agrobacterium-mediated transformation of 800 calluses of Zimoyarka variety with AGLO strain containing the pBi-OAT vector structure.
