Stefaniv I. Pharmacological characteristics of the new herbal medicine «Kasdent» for the treatment of inflammatory diseases of periodontium and oral mucosa

The dissertation of Stefaniv I.V. is devoted to the experimental study of the new herbal medicine, tincture «Kasdent,» created to treat inflammatory diseases of the periodontium and oral mucosa On a model of experimental gingivitis in rats, a 1:3 dilution with distilled water was chosen from three dilutions as the most effective breeding. The tincture «Kasdent»'s main pharmacological properties were determined: antiinflammatory, antioxidant, capillary strengthening, membrane stabilizing, and wound healing. The following models were used:gingivitis, chronic generalized periodontitis, and stomatitis. Under conditions of gingivitis, daily 4-5 times applications of the investigational agent for nine days slow down the inflammatory processes in the mucous membrane of the rats lower jaw: reduce the manifestation of clinical signs of inflammation by more than two times, restore hematological parameters (leukocytes - (14,75±0,56) 109 per l against (25,71±1,24) 109 per l in a group of positive control; sed rate - (1,50±0,18) mm/h against (4,0±0,92) mm/h), and reduce biochemical parameters (alkaline phosphatase by 2.7 times; acid phosphatase by 2.2 times; TBARS by 1.7 times; the level of reduced glutathione is increased 1.5 times). According to the results of microscopy, it was found that the tincture «Kasdent» helped reduce inflammatory changes and growth of the epithelial layer of the mucous membrane of the oropharynx. In chronic generalized periodontitis, tincture «Kasdent» in the treatment mode reduces inflammation intensity; this is evidenced by a decrease in signs of inflammation 2.6 or more times. The number of leukocytes is reduced by 1.3 and 1.5 times, sed rate -by 1.8 and 3.1 times. Tincture «Kasdent» on toxicological parameters at intragastric administration belongs to the VI class of toxicity (practically nontoxic substances). Also studied the safety of tincture «Kasdent» in dilutions of 1:2 and 1:3 with repeated injections in rats for 28 days. The investigational product does not cause significant violations of the functional state of animals' organs and systems, does not change the morphostructure of internal organs, does not cause irritation to the oral mucosa and the gastrointestinal mucosa of rats, does not show gastrotoxic and allergic properties. The research results open up prospects for further study of the medicine for implementation in industrial production and dental practice.
