Essay

The work is devoted to comparative study of absorbtion, distribution and excretion processes from the laboratory animals organism at different schemes of administration of [3H]-amixin and its metabolites. On the base of worked out analytical methods it was proved, that in the mice organism amixin is subjected to negligible biotransformation with two metabolites formation, with Rf values of 0,21 and 0,42. The amixin pharmacokinetics feature at its oral administration of introduction is a high rate of absorption in the organism inner medium and the low proccesses of its tissue and organ elimination. The processes of [3H]-amixine excretion from the mice organisms at all schemes of its oral administration during the whole time of experiment are realized with rather slow rate. The process of the total drug excretion with urine and feces – is nonparallel. The main route of elimination is excretion: of 39,2%with urine, and about 15% with feces of administrated dose. At the repeated amixine administration, the slowing down of the drug elimination processes is found, and isn’t manifested at single administration of the drug and should be accounted at course drug intake. Results of Mansgeldorf analysis method shows, that at infinite exposure about 80% of the administrated dose is excreted, at the single [3H]-amixin introduction on the background of the repeated administration of nonradioactive analog of drug and up to 52% is excreted after repeated introduction.
