Essay

The research deals with a screening study among monocarbamate derivatives for the purpose of searching a potential anticonvulsive agent. Using models of maximal electroshock and corazol seizures, anticonvulsive activity was established in 10 compounds among 32 new investigated carbamate derivatives. The following indices were established: median effective dose, minimal neurotoxic dose, therapeutic index, protective index of the compounds under study and that made it possible to single out the most optimal compound among them - coded AGB31. It has been shown that in the corazol kindling method, compound AGB31 prevents the development of EEG convulsive paroxysms of clinical convulsive states. It has been corroborated that a long-term administration of compound AGB31 does not lead to the development to its anticonvulsive effect. In addition to the anticonvulsive effect a moderate sedative and analgesic activity has been revealed in the neuropharmacological profile of the AGB31 action. It has been demonstrated that the mechanism of action of AGB31 is mediated by its GABA- and glycinergic effects. The obtained results of the research enable to consider compound AGB31 as a potential anticonvulsant agent.
