Essay

The thesis is devoted to an actual problem of the experimental pharmacology - to the research of substances with cerebroprotective effect, which can be further used for the creation on their basis a new medical product for treatment of an ischemic stroke. As it was established on the model of the acute distrubance of a cerebral blood flow (bilateral ligation of the general carotids) of the rats, the new aminoacid derivatives of 1,4- naphthoquinone are the carriers of the cerebroprotective activity. It became apparent by the lethality rate decrease among experimental animals at the preventive introduction of the studied substances (1-5% of their LD50) into their organisms. The greatest protective effect was revealed in the compounds containing a- alanine in their structure (compound 17) and glycine (compound 20). The mechanism of the cerebroprotective effect of the compound 17 is connected with its stimulating influence on the cerebral blood flow, as well as with its ability to improve rheological properties of blood by decreasing the content of fibrinogen in plasma, by blocking the activity tissue thromboplastin, and also having an anticonvulsant action.
