Essay

It is developed and optimized the system of methods of quantitative assay of new potential antihypoxant - derivative of propylthiobarbituric acid (compound I) and its metabolites in the biological substrates іn vіtro and in vіvo. It is shown that the compound I is subjected to extensive biotransformation in the organism of rats and mice with formation of 5-isopropylthiobarbituric acid and 5-isopropylbarbituric acid. With the help of outmodel analysis it is confirmed the rules and differences of biotransformation, distribution and excretion of parental compound and its metabolites that positively characterizes the its pharmacokinetic profile. The revealed peculiarities of the pharmacokinetics of investigated compounds at different species of animals can be used for more exact extrapolation of the obtained data on human organism. New methods of mathematical analysis of "rapid" processes of the compounds distribution in an organism, which allow to evaluate their role in pharmacokinetic processes and, thus, improve accuracy and validity of researches, are developed.
