Essay

The models with accute damage of the cerebral blood stream (bilateral ligature of carotid arteries) in non-druged rats, hypoxia of closed space and acute asphyxia found that derivatives of the 4-oxo(amino-)quinazoline are carriers of cerebroprotective and antihypoxical activity, which to a great extent belog to the compound 4-[oxo-3(4Н)-quinasoline]benzoic acid (PK- 66 – compound). Four day treatment of rats with accute damage of the cerebral blood stream (bilateral ligature of carotid arteries) by PK- 66 – compound (10 mg per kg intra musculular) as well as mexidol (100 mg per kg intra musculular.) promotes decrease in nevrological and cognitive deficits, easies expressions of energydeficits, normalises levels of oxidant-antioxidant equilibrium and protein oxidizing modification in ischemised cerebral cortex. Indicated changes in metebolistic processes are correlated with positive dynamics of the morphometric picture of sensomotorial cortex of the brain by the 21-st day after acute damage of the cerebral blood stream. This is expressed by almost complete renewal of neuron area and maintenance on them RNA, probable decrease in growth indicators of compactness neuroglia, amount of apoptosial and destructivly changed neurons on increase of index of neurons, which survived after ischemia in relation to untreated animals. Neiroprotective action of the 4-[oxo-3(4Н)-quinasoline]benzoic acid (PK- 66 – compound) is related to its capacity to stimulate cerebral gemodynamics, normalise metabolistic processses in ischemised cerebral cortex and existance of antihypoxial and cytoprotective efects.
