The thesis is dedicated to the investigation of the novel water-soluble derivatives (hydrochloride and megluminic salt) of (±)-cys-3-(2'-benzimidasolil)-1,2,2-trimethylcyclopentanecarbonic acid (Diacamph) cerebroprotective properties. Diacamph hydrochloride (10 mg/kg) is in the lead of the investigated compounds that exert an antihypoxic action as well as reduce lethality of rats underwent bilateral carotid occlusion against a background of normoglycemia and alloxan-induced diabetes mellitus. Diacamph hydrochloride is not inferior to mexidol (100 mg/kg) and surpasses the latter by several parameters. The links of Diacamph hydrochloride cerebroprotective activity are as follows: antihyperglycemic action, cerebral blood supply stimulation, antihypoxic properties, elimination of energy deficiency, metabolic acidosis and oxidative brain injury as well as maintenance of neurons integrity the mechanism of which includes antiapoptotic action (the reduction of DNA fragmentation in the nuclei of cortical neurons). Diacamph hydrochloride is a slightly toxic preparation (the ІV class of toxicity). The results experimentally substantiate the expediency of development and clinical investigation of diacamph hydrochloride injection preparation for the treament of acute cerebral ischaemia against a background of diabetes mellitus and normoglycemia.
