Essay

The thesis is devoted to experimental and theoretical substantiation of creating a new combinations of a non-opioid analgesics and antidepressants on the basis of researching of analgesic activity, anti-depressant properties and effects on behavioral responses in intact animals and in conditions of simulated depression. It is found, that in intact animals, the most effective analgesics were diclofenac, meloxicam, celecoxib, paracetamol; antidepressant activity had paracetamol (300 mg/kg), celecoxib (25 mg/kg) and diclofenac (10 mg/kg). Leaders in the analgesic and antidepressant activity in intact rats were combinations of meloxicam (2 mg/kg) and celecoxib (25 mg/kg) with amitriptyline (10 mg/kg). In depression analgesic effect of NSAIDs and their combination with amitriptyline was stronger, and the antidepressant - decreased, comparing to the effect in intact rats. The neurophysiological effects of the optimum combination of drugs and their components were also analyzed on the structures of the brain.
