Essay

The thesis is devoted to studying the effectiveness of diagnostic DNA-markers (to Is6110, katG, hsp65, hupB, rpoB, mpt64, mtp40) and genotyping markers (ETR-loci A, B, C, D, E, SNPs katG463 and gyrA95 and LSP deletion TbD1) on the clinical isolates of M. tuberculosis circulating in Kyiv among patients with pulmonary tuberculosis (PTB). Overall efficiency of PCR-diagnostics in sputum samples of patients with PTB was higher than of classical microscopic and cultural methods. Differential detection of PGG-1 and PGG-2/3 strains at the stage of early PCR-diagnosis with the SNP-specific marker katG is an informative approach for detecting the epidemiologically-significant PGG-1 strains, absolute majority of which are of the Beijing family in Kyiv's population of clinical M. tuberculosis strains. Genotyping of M. tuberculosis isolates using combined strategy of ETR-VNTR and SNP-typing slightly increases its accuracy and discriminatory ability. Strain families Beijing of PGG-1 and LAM of PGG-2 are the most common M. tuberculosis genetic variants and the most epidemiologically-significant M. tuberculosis families in Kyiv's region. Their further depth study is promising for improvement of strategy of M. tuberculosis characterization in the region.
