Essay

The thesis is devoted to experimental substantiation of expediency of coordination compound of germanium with potassium and citric acid (ОК-7) use for therapeutic purposes on a model of acute ischemic cerebral stroke. In the series of pharmakometric studies it was found out that the maximal anti-ischemic effect of the OK-7 is achieved in case of its intraperitoneal administration in dose of 64.92 mg/kg in 34.25 minutes after the start of acute cerebral ischemia modeling. It is shown, that in case of its single intraabdominal administration the OK-7 belongs to ІV class of toxicity ("low-toxic" agents). Pharmacological activity of the OK-7 in animals with acute cerebral ischemia is realized in "disconnection" of free radical chain reactions. No less important is the ability of the OK-7 to correct energy and carbohydrate metabolism disorders, normalize acid-base balance and enhance the natural detoxication ways aimed at reducing of endotoxins overproduction that ultimately ensures the preservation of the viability of the organism in acute cerebrovascular disease ischemic genesis. In case of stroke a significant modification of the OK-7 kinetics takes place. It is achieved by reduction of absorption rate, increase of deposition and slowing down the elimination, which in turn increases the presense of substance in the body and contributes to prolongation of its pharmacological effect.
