The thesis is devoted to theoretical and experimental substantiation of the feasibility and effectiveness of coordination compounds of germanium with nicotine and vine acids (ОК-3) for therapeutic purposes as means of cerebroprotection in the early posttraumatic period in closed head injury. When designing the optimal dosage regimen it was experimentally demonstrated that the maximum cerebroprotective effect of OK-3 is realized when it was administered intraperitoneally in a dose 119,2 mg/kg in 30 minutes and in dose 115,9 mg/kg in 7,18 hours after the pathology modeling. Cerobroprotective activity coordination compound of germanium, which was investigated, realizes in significant ability to prevent the development in dynamics of widespread membrane pathology in animals with closed traumatic brain injury by: the modification of free-radical reactions parameters, preservation of antioxidants profile components. Moreover, the potential cerebroprotector corrects the concentration of main components of adenyl-nucleotide system and level of energetic indexes, modifies the activity of main enzymes of energetic homeostasis and also normalizes the level of main parameters of carbohydrate metabolism under extreme conditions, which was modeled. The administration of OK-3 with a therapeutic purposes results in preservation components' activity of mitochondrial and microsomal electron transport chains of hepatocytes and normalization of functional state of livers' detoxicative system in closed head injury.
