Essay

The thesis is devoted investigation of structural and functional brain disorders on the model of prenatal alcohol intoxication and there pharmacotherapy by using cerebrocurin (0,06 mg/kg), tiocetam (125 mg/kg) and piracetam (125 mg/kg). Cerebrocurin and tiocetam revealed the most evident cerebroprotective action. The mechanism of energymodulating action of cerebrocurin is conditioned by its ability to limit development of mitochondrial dysfunction, promote expression of genes of the early reacting and expression of antiapoptotic protein bcl-2, thereby, activate the compensatory shunts of energy production that lead to increasing of neurons survival and as a result – preservation of brain cognitive functions. Antiapoptotic mechanism of neuroprotective action of tiocetam is due to the ability of its component (3-methyl-1,2,4-triazolo-5-thioacetate) to reduce early NO-initiation of apoptosis, acting as scavengers of cytotoxic wisecracking derivatives NO. In addition, tiocetam have capable of regulating protein synthesis and Red/Oxi-dependent expression of global transcription factors, that involved in the processes of memory and increase the speed of circulation of information macromolecules, protecting their structure from oxidative modification, shows energotropic action, and modulates the level of Bc1-2 in neurons.
