The thesis of Zeleniuk V.G. is devoted to the experimental rationale of statins as renoprotective agents under the conditions of acute renal failure (ARF). For the first time in four models of ARF was found the renoprotective action of several statins (atorvastatin, lovastatin, simvastatin), taking into account the duration and mode of drugs' administration, elucidation for their mechanisms of action, safety assessment, and identifying the best agent. The most pronounced renoprotective effectiveness of simvastatin (20 mg/kg) was proved in prophylactic and single dose administration, and atorvastatin (20 mg/kg) - after repeated administration within 7 days, and compared with reference drug Lipoflavon (296 mg/kg). Atorvastatin expressed increased glomerular filtration rate, and simvastatin - reduced content of protein in the urine, both - without causing unwanted myotoxic effects. New scientific evidence on the effectiveness of atorvastatin on renal function after the night and morning administration were obtained. It was demonstrated the ability of statins moderately affect renal function of intact rats, showing weak diuretic and antiproteinuric effects. It was established that statins are able to protect the kidney in experimental acute renal failure by prevention of an oxidative and nitrosative stress, recovery of an fibrynolytic and proteolytic activity, reduction of an endogenous intoxication and inhibition of an inflammation. Thus, the experimental results confirm the prospectivity of clinical trials of statins as renoprotective agents for the prevention and treatment of acute renal failure.
