Essay

The thesis experimentally proved the new approaches to reduce diclofenac sodium gastrotoxicity through the study of changes in the protective properties of gastric mucosa and the pharmacological effects of the drug in a different saturation of the organism with hydrogen sulfide. It was proved that the increase of H2S content in the body had a protective effect and increased the protective potential of gastric mucosa. It was established that the deficit of H2S increased, while the excess of it, on the contrary, reduced the degree of diclofenac sodium gastrotoxicity. The use of H2S donor in conditionally therapeutic dose (1/20 of the LD50) reduced the negative effect of diclofenac on stomach metabolom, the state of cell membranes and the contractility of mesenteric arteries, while the introduction of H2S synthesis inhibitor potentiated the effect of gastrotoxicity of such NSAIDs. NaHS showed antiapoptotic effect and had a strong corrective action on changes in the indices of gastric mucosa cell cycle caused by diclofenac sodium.
