Essay

The thesis is devoted to theoretical and experimental basis of search of 2-oksoindolin-3-glyoxylic acid substances with antidepressive activity and also definition of their possible mechanisms of the action. It was detected the most active substance (ethylic ether 4- [2-hydroksy-2-(2-okso-1,2-dihidro–indol-3-iliden)-atsetamino]-oil acid of E-38 with antidepressive action based on complex analysis of emotional and behavioral tests. E-38 increased dopaminergic neurotransmission and also decreased haloperidol catalepsy and intensified effects of apomorphin and L- dihydroxyphenylalanine. There were established serotonergic and noradrenergic components of antidepressive action of E-38 at reserpine and clonidine depression. There were shown mediator mechanisms of antidepressive action of derivative of 2-oksoindolin, that correlate with the increase of dopamine level, noradrenaline level and serotonin one and normalization of monoamine oxidase A activity.
