Essay

It was established that a single intraperitoneal introduction of new derivatives (3-R-2-oxo-2H- [1,2,4] triazine [2,3-C] quinazoline-6-yl) alkylcarboxylic acid (10 mg/kg) (except DSK-70, DSK-106, AV-225 and AV-229), as 2-ETBI (bemityl, 50 mg/kg), to rats leads to increasing of swimming test duration, which may be a sign of their actoprotective activity. This effect was the most in sodium salt 4-(2-oxo-3-phenyl-2H-[1,2,4]triazine[2,3-cquinazoline-6-yl)butanic acid (DSK-38). DSK-38 demonstrated actoprotective effect in normal and difficult conditions of the experiment (hypo- and hyperthermia, hemic and circulatory hypoxia, experimental myocarditis, hypokinesia). It was the same and sometimes prevailed reference aсtoproteсtor 2-ETBI. Courses of DSK-38 15-day administration (4,8 mg/kg, i/p) as 2-ETBI (34,0 mg/kg, i/p) to hypokinetic rats lead to decrease of the destructive processes and optimization of adaptation processes in skeletal muscles and internal organs of animals. The mechanism of actoprotective action of DSK-38, as 2-ETBI, consists of antihypoxic effect, the ability to stimulate synthesis of RNA and DNA, to eliminate deficiency of energy, to store carbohydrate reserves and to normalize prooxidant-antioxidant balance in the blood, liver and skeletal muscles of rats.
