The thesis is devoted to the complex pharmacological study of the firstly synthesized 7,8-disubstituted derivatives of theophylline. It has been established that the majority of the studied compounds exhibit high diuretic, analgesic, anti-inflammatory, antihypoxic, and antispasmodic activities, not yielding to, or even surpassing, the effects of known comparator drugs. First identified the leading substance − 7-n-methylbenzyl-8-n-bromobenzylidenhydrazinotheophylline (code name benofillin). In two experimental models of acute kidney damage induced by ethylene glycol and glycerol, benofillin has been shown to eliminate impaired water-electrolyte metabolism, improve the nitrogen-releasing function of the kidneys, suppress free radical oxidation processes in the kidneys, and restore their morphostructure more effectively to canephron and chophytol. The established efficacy, low toxicity and the absence of an ulcerogenic action substantiate the possibility of creating, on the basis of this compound, a new diuretic with nephroprotective activity.
