Essay

The thesis deals with the experimental study of toxic and pharmacological properties of a new nanocompound – nanochromium citrate (NCC). The parameters of NCC toxicological characteristics are clarified, the main pharmacological properties are found with establishment of “dose-effect” dependence. In case of intracutaneous (i/c) administration NCC belongs to ІІІ class (moderately toxic substances), and in case of intravenous (i/v) injection – to ІІ class (highly toxic substances). Dose-dependent effect of NCC is detected on the function of the kidneys, liver, cardiac and systemic hemodynamics, and oxidative/anti-oxidative balance. For the first time hypoglycemic activity of NCC is found with different ways and regimes of administration. Inversely proportional relations exist between the dose and hypoglycemic activity. On the model of experimental diabetes mellitus (EDM) anti-hyperglycemic activity, influence on insulin content, insulin resistance index, and hypolipidemic effect of NCC were not found to be inferior to the reference drug metformin with an increasing protective on the histological structure of the pancreas. NCC possesses membrane stabilizing action (increases osmotic resistance of erythrocyte membranes). Thiotriazolin efficacy is evidenced in case of NCC toxic action (decreases pro-oxidant activity, increases survival, reduces mortality rate of animals).
