The thesis is devoted to the theoretical and experimental substantiation of the feasibility of the use of metadoxine as a detoxicative and hepatoprotective agent to prevent the development of acute alcohol intoxication and liver injury of alcoholic and non-alcoholic etiology. Metadoxine prophylactic administration to animals on the background of the action of the acute alcohol intoxication factor accelerates the recovery of reduced spontaneous locomotor activity, reduces the frequency of muscle relaxation, the urge for vomiting, prevents the impairing of cognitive performance and accelerates the renal elimination of ethanol. Metadoxine prophylactic administration on the background of the action of alcoholic or non-alcoholic liver damage factor doesn’t completely prevent the metabolic liver damage, but significantly reduces the severity of the liver biochemical abnormalities. According to the assessment of the possible ionized/non-ionized forms of metadoxine components and in vitro dissolution kinetics of immediate release solid oral dosage forms of metadoxine with systemic action in the physiological pH range, it is appropriate to administer metadoxine tablets 30–60 min prior to administration of acute alcohol intoxication and toxic liver damage factors.
