The thesis is devoted to theoretical and experimental substantiation of expediency of purposeful synthesis and search among 4-oxo (amino-) quinazoline derivatives of compounds with analgesic and anti-inflammatory effects, promising for creation on their basis of a new non - narcotic analgesic, competitive with ketoronolacrum trotamate. In screening studies of the analgesic action of 4-oxo (amino-) quinazoline derivatives in models of pain caused in rats by electrical, thermal and chemical factors, a compound of the leader 4- [4-oxo- (4H) -quinazolin-3-yl] benzoic acid (PK-66), competitive with diclofenac and ketorolac. An in-depth study of the pharmacological properties of PK-66, and its effect on the components of inflammation. The greater safety of the compound - a leader in the gastrointestinal tract at 30 days of intact rats, compared with modern non-narcotic counterparts. The therapeutic effect and safety of PK-66 on the AA model were evaluated. The main neurochemical and mediator mechanisms of antinociceptive action are established and the mechanism of anti-inflammatory effect of the leader compound is characterized. The prospects of PK-66 for its creation on its basis of new NNA or NSAIDs, competitive with ketorolac tromethamine and diclofenac sodium, have been determined
