The dissertation is devoted to biochemical mechanisms study of metabolic abnormalities in proteins, carbohydrates, lipids, free radical lipid peroxidation condition and thyroid hormones metabolism under pathological conditions, related to hyperthyroidism in experimental animals and clarifying mechanisms of newly synthesised 2-oxo-4-hydroxychinoline derivatives correction influences on the mentioned processes in comparison with mercasolyl. Under hyperthyroidism model pathologies (thyrotropic, thyroxin, cold) and abnormal thyroid gland patients study high antithyroid efficacy of 1H-2-oxo-3-(-(benzimidazol-2-il)-4-hydroxychinoline derivative named "tetrakon" was found. The biochemical mechanisms of specific tetrakon activity were studied, which are related to thyroid hormones formation blockage and their elimination inhibition from thyroid gland tissue. Extrathyroid mechanisms of correction tetrakon activity on protein, carbohydrate, lipid metabolism, tissue respiration and energy formation processes andrelated to the mentioned above protective activity on functional state of liver in hyperthyroid animals were determined. Hypertrophic side effects absence in tetrakon is proved by thyroid gland morphological structure restoration in rats with hyperthyroid condition. For 2-oxo-4-hydroxychinoline derivatives as perspective thyrostatics advantages over well-known drug mercazolil are defined.
