Essay

Object – neuroretinoprotection mechanisms for ischemic and traumatic injury of the visual analyzer; aim – establish the role of NMDA receptor modulators in the mechanisms of neuroretinoprotection in ischemic and traumatic eye damage, and on this basis experimentally substantiate new approaches to correcting the pathology of the visual analyzer; methods – biochemical, enzyme immunoassay, functional, flow cytometry, pharmacological, statistical; results –the role of pharmacological modulators of different sites of NMDA receptors in the mechanisms of neuroretinoprotection under the ischemic and traumatic damage of the visual analyzer in vivo experiments has been determined; the feasibility of correction of neurotrophic-destructive changes in ischemia / reperfusion of the eye and contusive eye injury with the help of the inhibitor of the amine diamine sulfate polyamine site and the inhibitor of the phencyclidine site of memantine was experimentally substantiated; the lower efficiency of the blocker of the ion channel of magnesium sulfate was proved; for the first time there were shown that under ischemic and traumatic damage of the visual analyzer, an increase in the level of NSE and S100 protein in blood serum was associated with the formation of an unfavorable metabolic pattern, and the increase in the cytometric signs of apoptosis and neuroglioproliferative activity in the retina at different times took place; it has been established that in case of ischemic and traumatic damage of the visual analyzer, neuroretinoprotective effect of NMDA receptor inhibitors is realized not only through the modulation of glutamate excitotoxicity, but also because of a decrease in the retina of microcirculation disorders, a decrease in the level of proinflammatory and proaptotic factors, an increase in the activity of glutathimeter therapy, and an increase in the activity of glutathimeter therapy; for the first time, there was proved that amantadine sulfate significantly excels memantine in its ability to accelerate the de-escalation of the NSE level and S100 protein in the blood, to correct cell cycle parameters (with a decrease in the level of DNA fragmentation) and on the metabotropic effect; branch – medicine.
