Essay

The thesis is devoted to the experimental solution of the problem of analgesia and neuroprotection in the experimental modeling of multiple sclerosis. Analysis of antinociceptive activity spectrum shows that in the conditions of demyelinating disease (experimental allergic encephalomyelitis) during basic hormone pharmacotherapy meloxicam should be considered as a drug of choice for the correction of somatic pain, which showed a high level of analgesic effect and significantly reduced the activity of PGH-synthase in the brain and spinal cord. Pharmacological analysis of antinociceptive and neuroprotective activity of the combined use of classical and adjuvant analgesics and the drugs for brain protection with methylprednisolone as the drug of basic hormone pharmacotherapy in conditions of experimental equivalent of multiple sclerosis allowed to determine the effective and efficient direction of pharmacotherapy of pain syndrome through the use of combinations of "methylprednisolone-meloxicam" and "methylprednisolone-gabapentin", which contribute to the preservation of sufficient antinociceptive potential of meloxicam and gabapentin. Also this allowed to establish strategic direction of neuroprotective therapy of experimental allergic encephalomyelitis due to combination "methylprednisolone-citicoline" that contributes to the inhibition of the biodegradation of sphingomyelin and cardiolipin, restores the activity of glutathione reductase, stimulating glutathione synthesis and exhibiting anti-apoptotic effect.
