Essay

The dissertation describes the experimental ground for use of fibrous and granular carbon enterosorbents alone and in combination with hematopoietic cytokine to ameliorate the side effects of anti-cancer drugs. Bone marrow suppression, gastro-intestinal toxicity, impairment of fertility and damage of detoxifying organs and tissues are the most frequent side effects of classical dose dense and dose intense anti-cancer chemotherapy. Hematopoietic cytokines as granulocyte colony stimulating factor G-CSF are the part of conventional therapy in this field, but their effects require boosting. That is why the use of means and methods of adsorption therapy (as a part of Sorption Detoxification) is considered promising. Use of enterosorbent or combined use of both preparations provided significantly better effects toward the prooxidant/antioxidant balance in rats. But, the combination was the most efficient to restore the balance in the pro- and antioxidant system. Filgrastim alone promoted a slight, yet significant improvement of the indices only in the liver tissue, namely TBARS, SOD and G-SH, while the combination of G-CSF with enterosorption caused normalization of all indices and even more. Enterosorption ameliorates side effects of subchronic doxorubicin toxicity. We observed improvements of hematological parameters, kidney’s function and decrease of endogenous intoxication markers. But the combination of enterosorbent C2 and G-CSF demonstrates more significant enhancement of tests, especially activity of AST by 26.3 %, CPK-MB by – by 17.0 %; ALT – by 20.4 %. Enteral sorption therapy mostly normalized and improved violated indices and decreased the rate mortality of rats. We observed also shrinking of the endocardial surface of the left ventricular wall area by 21.3% and decreasing of Planimetric Index. Those results demonstrate that enterosorption could prevent remodeling of the heart chambers.
