Kostenko K. Search for substances with anorexic and anxiolytic properties among 3-substituted 1,4-benzdiazepine-2-one and 1-arylpiperazines derivatives

Dissertation is dedicated to the experimental search for the potential medicinal agents with anorexic and anxiolytic action among new derivatives of 3-amino-1,4-benzodiazepine-2-one and 1-aryl-4-[4-(N-naphthalimido)alkyl]piperazines. 36 substances were studied, they are related to two different classes of chemical compounds with different substituents. In result of pharmacological investigation of new synthesized compounds on appetite, presence of anorexic and orexic activity in these agents was revealed in low doses 0,07-2,0 mg/kg. These compounds may be used as potential agents for treatment of anorexia, obesity, bulimia as well as panic and anxious states. It was shown the anorexic activity of 1-aryl-4-[4-(2-naphthalimido)alkyl]piperazines depends mostly on their structure - on the length of methylene chain as well as on the presence of various substituents in phenylpiperazine and naphthalimide rings. It has been established that given compounds are ligands of cholecystokinine CCK2-receptors.
