Essay

In present work the research of resistance of M.tuberculosis to antituberculosis agents in Odesa region, and capability of polymerase-chain reaction (PCR) methods for detection and following-up of resistant strains was fulfilled. It was established that in 2006 y. among the first- and second-line antituberculosis agents the lowest resistance was for pyrazinamide and pefloxacine, while the highest one – for rifampicin and ofloxacine. Multiplex allele-specific PCR method revealed the highest specifity for mutations in rpoB gene and the highest sensitivity for mutation in katG gene, which are responsible for rifampicin- and isoniazide-resistance. Presence of katG and rpoB mutations has been more frequently observed in ex-detainees and at repeated course. Also presence of these mutations associated with lower effectiveness of tuberculosis treatment. According to the results of 6 loci detection the high level of katG і rpoB genes’ mutations has been observed in a number of clusters and groups of M.tuberculosis. The obtained results proved importance of monitoring of the drug-resistance for M.tuberculosis, witnessed possibilities and importance of PCR-method for detection and identification of resistant strains of M.tuberculosis.
